NAD+ and Bipolar Disorder: What the NR Research Means
Why brain-energy research and a study at a bipolar center do not establish NAD+ treatment for bipolar disorder, mania prevention or mood stabilization.

NAD+ injections, IV therapy and oral NAD precursors are not established treatments for bipolar disorder. The distinction between research on brain energy and treatment of mood episodes matters here. A study location or laboratory outcome can easily be mistaken for clinical treatment evidence 1.
Our NAD+ and depression evidence guide covers broader mood claims. This article addresses a different question: whether evidence applies to bipolar depression, mania or maintenance treatment. The NAD+, NMN and NR comparison explains why a precursor capsule and an NAD+ infusion also need separate evaluation.

NCT07649161 lists Massachusetts General Hospital as sponsor and the Dauten Family Center for Bipolar Treatment Innovation as its location. But its registered condition is Healthy Adult, with no current psychiatric diagnosis or first-degree family history of mood or psychotic disorders allowed 1.
The plan is an open-label, single-group oral NR study with an estimated 50 participants. Its primary outcome is the brain NAD+/NADH ratio measured by magnetic resonance spectroscopy before and after two weeks. The registry had no posted results at our October 15, 2026 review; its last update was posted June 17, 2026 1.
Those details define what a result could mean. Even if the ratio changes, that alone cannot show that an intervention relieves bipolar depression, prevents mania or reduces future episodes. Those questions require studies in the relevant patients with clinical outcomes.
| Research | What it can tell us |
|---|---|
| Healthy-adult NR study | Brain NAD+/NADH measurement; not a bipolar symptom trial. |
| Other-population mood outcomes | Eligibility restrictions limit application to unstable bipolar illness. |
| Clinical bipolar treatment claim | Needs evidence for the relevant mood phase, intervention and patient outcomes. |
Does a biomarker show that mood is improving?
A measurement can be useful for demonstrating that an intervention changes a biological process. It does not automatically become a validated measure of a patient's recovery. The NAD+ therapy evidence guide explains this distinction across the field.
For bipolar disorder, a clinically useful claim would need to specify the phase being treated and the outcome. Relief of a depressive episode, prevention of a manic episode and maintenance of long-term stability are separate questions. A favorable result on one would not establish the others.
There is another gap between oral NR and a clinic's NAD+ infusion. A claim about an infusion needs evidence for that actual intervention; it cannot inherit a treatment effect from a precursor study or from a brain scan. Our NAD+ IV evidence review covers the route separately.
Why trials in other populations cannot settle this
An NR trial in people with long COVID measured depression and anxiety among its outcomes. Its eligibility criteria excluded major active or chronically unstable psychiatric illness, including bipolar disorder, within the previous year 2.
That restriction matters even if an advertisement describes the work simply as “human mood research.” A study that excludes the psychiatric population of concern cannot establish a bipolar treatment regimen or prove safety during a manic, mixed or unstable depressive episode. It also does not answer how NR interacts with a person's specific mood-stabilizing medicines.
The correct reading is neither that every mechanism is meaningless nor that any human trial proves the clinic's claim. Check who participated, what they received, the comparison group and the actual outcome before applying a finding to your own diagnosis.
Could feeling more energized be a warning sign?
More energy is not always the same as better mood stability. NIMH lists decreased need for sleep, racing thoughts, unusually increased activity and feeling wired among symptoms that can occur during mania. Bipolar disorder can involve depressive, manic or hypomanic episodes and sometimes mixed features 3.
This does not establish that NAD+, NR or NMN causes mania. The sources reviewed here do not quantify that risk. It does mean that a new period of very little sleep with unusually high energy deserves clinical attention, even if it follows something marketed for wellness. Avoid assuming that every activating change is a benefit or, conversely, that timing proves the supplement caused it.
If these changes occur, contact the treating mental-health team promptly and describe the symptoms, timing and complete product ingredients. If there is immediate danger, seek emergency care. Do not wait for a supplement experiment to establish the explanation.
What to discuss before adding a product
NIMH describes long-term treatment with medication and psychotherapy as central to managing bipolar disorder. It advises continuing treatment and discussing changes with a health-care provider 3.
Bring the complete label and current medicine list to the prescriber or pharmacist. Ask whether the proposed add-on has been studied in the same phase of bipolar disorder and alongside the same medications, what benefit is being targeted, and how emerging sleep or mood changes would be handled. Our NMN interaction guide explains why a lack of published interaction data cannot confirm that a combination is safe.
There is no validated bipolar NAD+ schedule to extract from the research discussed here. A healthy-adult brain-energy protocol is a research tool, not a replacement for a treatment plan designed to keep mood stable.
Frequently asked questions
Is the NR study at the Dauten bipolar center a bipolar treatment trial?
It is a healthy-adult brain-energy study. Its design cannot establish treatment benefit in bipolar disorder.
Can NAD+ or NR replace a mood stabilizer?
The evidence reviewed here does not establish that role. Discuss treatment changes with the prescriber instead of substituting a wellness product.
Does NAD+ cause mania?
The sources reviewed here do not quantify a mania risk from NAD-related products. New decreased need for sleep, racing thoughts or unusually increased activity warrant prompt clinical review regardless of the cause.
